The former Lost actor will play Tyler Martin, an English rock star who plays an instore gig at Buy More and gets caught up in one of Chuck's spy missions.
Speaking to Entertainment Weekly, Monaghan acknowledged that the character was similar to his Lost rocker Charlie Pace.
He said: "Obviously there are similarities [to Charlie]. But this is a comedic role. Charlie was not incredibly comedic on purpose. I think Charlie had funny moments, but Charlie was quite tortured and into meth.
"The guy that I play on Chuck is just an out-and-out lunatic. If you were to look up 'definitive rock star' in the dictionary, there would be a picture of him."
Monaghan added that he agreed to take on the role for the chance to work with Chuck co-creator Josh Schwartz.
"I've been wanting to be in business with him for a long time," he said.
Monaghan's guest spot will air on February 2.
