The channel's entertainment president Robert Greenblatt confirmed that the offshoot is set behind bars and likened it to "a female Oz" but "not as dark".
Greenblatt added that the pilot's three guest stars, Melissa Leo, Famke Janssen and Laurie Metcalf, may return as regulars should the pilot go to series.
The last season of The L Word, airing in the US this Sunday, begins with the death of Mia Kirshner's Jenny before flashing back to reveal the identity of the killer.
Asked if Alice's prison sentence means that she is the murderer, series creator Ilene Chaiken said: "That's a complicated question. The two [shows] stand alone separately, but should the spinoff get picked up as a series, then yes, it will."
