The actress played Nurse Carol Hathaway in the NBC medical drama from 1994 until 2000.
When asked by Us Weekly if she would return, Edwards said: "Of course! I have nothing to base that on. But I think it will happen."
Marguiles's character eventually left ER to live in Seattle with Dr Doug Ross, played by George Clooney.
The Ocean's Eleven star was photographed on the show's LA set last month, appearing to confirm rumours that he would be making a special appearance in the show's final season.
"I'm happy for the show. Everybody who was there in the original cast has all come back and showed up, and it's nice thing," added Edwards, who played Dr Mark Greene.
"I think it shows it's a group who are appreciative of what was given to them."
The final episode of ER will air on April 2 and will be preceded by a one-hour retrospective.
