Bigger, Longer & Wider will be her third time presenting a comedic programme for the TBS network, TV Guide reports.
She said: "I love Chicago, and if it weren't for the weather, I'd live there. And you know I've performed many times in the [Chicago Theatre], so it's fun to go back to that theatre. I kind of started there."
The special will feature guest stars Kanye West, Nick Cannon and magician David Blaine, while she also explained that she loves the idea of hosting something other than her daily talkshow.
She added: "I have fun doing [variety shows] - it's just something different to do than my show every single day. I grew up watching Rowan and Martin's comedy hour, Carol Burnett, Sonny & Cher and Donny & Marie.
"I loved those shows. So it's just something I've always been drawn to, and it kind of has something for everybody."
DeGeneres was recently nominated for two Daytime Emmy awards.
