The Superman Returns actor will be featured in a multi-episode story arc on the show, Entertainment Weekly reports.
The 29-year-old will portray an enigmatic new agent named Shaw, who will serve as a mentor to the titular character (played by Zachary Levi) and compete with him for the affections of fellow spy Sarah (Yvonne Strahovski).
Routh will appear in the fourth episode of the new season, which begins in March 2010.
It was recently announced that Routh is no longer under contract to reprise his role as The Man Of Steel.
