The former Saved By The Bell star and current Extra host will serve as master of ceremonies for the live show on January 30.
It will take place at the Planet Hollywood Resort and Casino in Las Vegas and will feature 53 contestants, including Miss Puerto Rico who is making her return to the show after a 50-year absence.
Lopez previously helmed the event in 2007 and 2009, which saw Indiana's Katie Stam win the honour.
