The Spider-Man creator will play himself in the March 1 instalment of the CBS sitcom, reports TV Guide.
The main characters in the show, played by Johnny Galecki and Jim Parsons, will run into Lee at their favourite comic book shop, Kapow!
Lee has appeared in several films based on his Marvel comic book characters, including Iron Man, The Incredible Hulk and Fantastic Four.
The Big Bang Theory airs Mondays at 8:30pm ET on CBS.
