Wheaton's first guest appearance on the CBS comedy helped boost its ratings by 15% in the 18-34 demographic last October.
According to TV Guide, the actor will return as an evil version of himself and nemesis to Sheldon (Jim Parsons).
"Even though my personal motto is 'Don't Be A Dick', I've wanted to play an arch enemy for pretty much my entire career," he posted on his personal blog.
Wheaton added that he will tape his spot on March 2.
