The show has struggled in the ratings since debuting on September 21.
Arnett wrote on his Twitter feed: "Never done a grassroots effort, but let's save the effing show Running Wilde forever! Or [at least for] five decent seasons."
The actor also joked about the comedy's falling viewing figures.
He continued: "Thanks to everyone who signed the petition. F**king ratings sucked. I guess America hates jokes?"
Arnett's co-star David Cross previously expressed his shock at the negative reviews the series has received.
Running Wilde continues on Tuesdays at 9.30/8.30c on Fox.
