The comedy focuses on a young couple called Zack (Kyle Howard) and Gillian, who feel "smothered" by their very different sets of parents.
Deadline says that McGinley, who is best known for his role as Dr Cox in Scrubs, has signed up to play Zack's father. He will appear alongside Marcia Gay Harden, who will star in the project as Zack's mother.
Meanwhile, Arkin will play Gillian's dad alongside Julie White, who has already landed the role of her mum.
Arkin has previously starred in a number of shows including Sons Of Anarchy, Life, 8 Simple Rules, The West Wing, Chicago Hope and Northern Exposure.
Smothered has been created by Friends producers Andrew Reich and Ted Cohen.
