The actor told Entertainment Weekly that he will play a "sick character" in an April episode.
"I love it," he said. "I'm having such a good time. It's just such a different role form me. But when you see it, you go 'Oh, that makes perfect sense'. The casting is really kind of an interesting choice to put me in there."
The former ER star also admitted that he is relieved to be taking a break from more light-hearted roles.
"I've been doing lighter stuff [like] Glee and some funny stuff on Entourage this year," he said. "It's been really nice to get to dig into a darker character."
Stamos will appear alongside Footloose actress Lori Singer in the instalment.
Law & Order: SVU airs on Wednesdays at 10/9c on NBC.
