The pilot, which has been penned by Sex And The City scribe Michael Patrick King, stars Don Johnson as an ageing Beverley Hills hairdresser called Allan Mann who is determined to stay relevant.
Deadline says that Barkin has now signed up to play Allan's ex-wife and the mother of his children.
The character works as Allan's financial adviser but she also gives him emotional support.
Barkin has previously starred in movies including Fear and Loathing in Las Vegas, The Big Easy, Drop Dead Gorgeous and Brooklyn's Finest. A Mann's World would be her first series role.
A Mann's World also stars Mario Cantone, Sandra Vergara, Kelly Hu, JR Bourne and Taylor Kinney.
