The actress, who has previously appeared in sitcoms Cheers and Veronica's Closet, will play the lead role in The Manzanis, a multi-camera family comedy written by Marco Pennette and Dave Flebotte.
The sitcom is described as similar to 1990s series Roseanne, with Alley starring as a wife and mother whose family moves to a "repressed country-club town".
The 60-year-old finished as runner-up on ABC's Dancing with the Stars in May.
Alley had previously hinted that she had two sitcom projects in the works in April, although she elaborated that the sitcoms were for TV Land and HBO.
