The cable network has ordered an untitled comedy starring Nathan Lane, as well as the series Paging Dr Freed, according to The Hollywood Reporter.
The order comes as part of an attempt to expand the network's entertainment brand.
"The pick-up of these half-hour comedy pilots underscores our commitment to broadening the appeal of USA's entertainment brand," said the network's co-presidents Chris McCumber and Jeff Wachtel.
"We're excited to be collaborating with some of the most distinct voices in the business as we open up to new genres."
Paging Dr Freed centres on two brothers who inherit their father's medical practice after he dies. Meanwhile, the Nathan Lane project follows an unfortunate actor who returns home to Texas when his father's health begins to deteriorate.
In August, USA renewed the legal drama Suits for a second season. In addition, the comedy-drama White Collar will return for a fourth season in 2012.
