In Little Brother, Stamos will play a man who discovers he has a half-brother who happens to be an ex-convict, reports Deadline.
The ex-con half-brother will be played by comedian T.J. Miller, whose previous roles include the 2010 movie Gulliver's Travels and ABC's Happy Endings.
The pilot is written by Everybody Loves Raymond's Mike Royce and will be shot by Real Steel director Shawn Levy.
Last month, Stamos was said to be circling Little Brother. He was also involved in rewriting the pilot with Royce and Levy.
