The actor told Ryan Seacrest on KIIS-FM that reports of Jennifer Aniston playing a guest role were "a thousand percent not true".
"You sort of want a show to get its sea legs and for you to understand that these are new people that you're trying to introduce," Perry explained. "I think it would be confusing if someone from Friends was on right away."
Go On stars Perry as a sports journalist who enters group therapy to overcome the loss of his wife.
The 42-year-old previously admitted that he is open to the Friends cast appearing on the show at some point in the future.
"It would be fantastic to get the guys on the show," he said last month. "Not right at the beginning though. But further down the line? That would be fantastic. I'd love that."
NBC will air a special preview of Go On on Wednesday, August 8, with the series debuting in full on September 11.
