The actress - known for playing Ari Gold's wife on the HBO series - has been cast as Danielle Deaver in The Secret Lives of Husbands and Wives.
Produced by Jerry Bruckheimer, the pilot will focus on a murder and how it complicates the personal lives of three suburban couples and their families, Deadline reports.
Reeves's character Deever is described as "flirty and funny" and is said to be hiding a dark secret. The Secret Lives of Husbands and Wives has been written by Sascha Penn and is based on Josie Brown's 2010 novel of the same name.
In addition to her Entourage role, Reeves has appeared in recent episodes of NCIS and White Collar.
Other pilots currently in the works at NBC include a new comedy from Community star Donald Glover, a sitcom produced by Jon Favreau and a legal drama starring Angela Bassett.
