The ABC TV project is loosely linked to Whedon's movie The Avengers - one of several films in which Jackson has played Marvel Comics hero Nick Fury.
"I keep hearing about it, and I haven't talked to Joss about it," the actor told MTV.
"But in my mind, I would tend to think that he would, if nothing else, be like Charlie in Charlie's Angels - at least [appear as] the voice!"
Marvel TV's Jeph Loeb has also revealed that S.H.I.E.L.D. could debut as a full series late next year.
"We have a 'little' pilot for television that you might have heard about called Marvel's S.H.I.E.L.D. co-written, directed, and executive produced by none other than Joss Whedon!" Loeb told PREVIEWSworld.
"[We're] bringing in Clark Gregg to play Agent Phil Coulson - the best! Hopefully we will be on in the fall of 2013 on ABC!"
ABC president Paul Lee recently admitted that the network is "very excited" about Whedon's pilot.
"We're optimistic, because Joss is a genius and we love the idea and we love what we've seen," he said last month. "It's a long way to go between now and a successful series, but we are very excited about it."
