Blair (Cruel Intentions) is in talks with the network to play Kim while Molly Shannon (Will and Grace) has agreed to take the role of Kath.
NBC bought rights to develop the format a year ago but it has been delayed repeatedly.
Kath and Kim creators Jane Turner and Gina Riley, who also played the main characters in the Australian original, will act as executive producers on the US pilot.
They will be joined by writer Michelle Nader while Reveille and Universal Media Studios will co-produce.
The show has aired in the UK on BBC Two.
