The rapper will make an extended cameo across two episodes of the ABC programme, to air on May 8 and 9.
He will sing tracks 'Sensual Seduction' and 'Life Of Da Party' from his ninth studio album Ego Trippin', as well as remix the show's theme song.
He said: "I've been a fan of One Life to Live since I was a baby. My momma always had it on the tube in the crib growing up.
"The opportunity to change up the theme song and give it some of my flavour will make the show the Life Of The Party."
Singers including Mary J. Blige, Nelly Furtado, Timbaland and Keri Hilson have also performed on the soap.
An animated comedy series about the rapper's early life is being developed in the US.
