According to E! Online, producers want the 20-year-old to play Annie Mills, an emo-mad music and theatre student.
It was recently reported that Rob Thomas, who wrote the draft script for the new show, is keen for Jennie Garth to return as Kelly Taylor.
Tori Spelling, who played Donna Martin throughout the programme's ten-year run, has also said she would love to appear.
The CW show will follow a new generation of youths living in the wealthy suburb featured in the 1990s series.
Duff has launched a successful film, fashion and music career since leaving Disney series Lizzie Maguire in 2004.
