The Smallville star will play Samantha Masters, a romantic interest for Bosley (Ramon Rodriguez), according to TV Line.
A tough CIA agent, Masters will reunite with her former love when the pair are both sent on assignment to Cuba.
Durance is best known for playing Lois Lane on Smallville from 2004 to 2011, and has also appeared in episodes of True Calling and Stargate SG-1.
The actress also recently signed up to star in medical drama pilot Saving Hope for Canadian television network CTV.
Charlie's Angels executive producer Al Gough recently revealed that Durance's former Smallville co-star Michael Rosenbaum is keen to provide the voice of Charlie on the show.
At the time, Gough said: "I think he might be a little young, but we'd obviously love to have any of [the Smallville alums] come on the show at some point."
Charlie's Angels premieres on Thursday, September 22 on ABC, and will air on E4 in the UK. Durance's appearance is expected to air in October in the US.
