The project will focus on a group of aspiring performers who come together to stage a musical based on the life of Marilyn Monroe.
Deadline reports that Jaffrey will play the boyfriend of the musical's lead actress, Karen (Katharine McPhee).
The actor previously starred in BBC drama Spooks from 2004 to 2007 and recently appeared in an episode of NBC's The Cape.
Other actors confirmed to appear in Smash include former Will & Grace actress Debra Messing and The Bounty Hunter star Christian Borle.
The pilot is under consideration for NBC's 2011-12 television season.
