The 29-year-old, currently best known for his part as Walford's Dr Al Jenkins, is to play Baldur, the Norse god of light on the CW series.
News of Croasdell's casting comes just a few days after his final EastEnders scenes aired on BBC One. He had been out in Los Angeles for only a week when he secured his new part.
Speaking of the role, Croasdell told DS: "It's fantastic being a part of such a popular show and exciting to be in the American arena. I'm looking forward to the challenge."
The soap star had previously confirmed talks for new projects by revealing that he had "a couple of nice things on the boil", but kept tight-lipped over the details at the time.
As well as EastEnders, Croasdell has appeared in The Chase and Holby City. Meanwhile, last November, he revealed that he had taken on the role of James Bond for Activision's latest game.
Supernatural's current fifth season is expected to be its last, though creator Eric Kripke has suggested that there is room for another season if the show is recommissioned.
