In the recent finale, Dean (Jensen Ackles) returned to Lisa and Ben in an attempt to live a normal life.
Kripke has now told Entertainment Weekly that the new episodes will centre on Sam (Jared Padalecki) and Dean being "on the road together".
However, he added: "Dean's situation causes all sorts of angst and conflict. The question [we'll be] exploring is, 'How can you be a hunter when there are actually people that you care about?' I think we'll mine it for all of the emotional angst and conflict that we can.
"But [Lisa] is not - nor is anyone - going to be riding in the backseat of the Impala with Sam or Dean."
Supernatural returns for a new season on September 24.
