It was reported last month that a forthcoming episode of the CW drama would poke fun at the vampire franchise.
"The fans of Supernatural are pretty cool," Ackles told Zap2It. "They're not crazy Twilight fans. They actually have some self-respect. I think we're lucky in that aspect."
The actor - who plays Dean Winchester - also revealed that he had received some unusual gifts from viewers since the show began in 2005.
"I've gotten some pretty interesting presents in the mail," he admitted. "God love them, my parents receive stuff."
He joked: "Don't send my parents stuff. Especially spells... [with] used underwear."
The sixth season of Supernatural begins September 24 on The CW.
