She told TV Guide that a number of popular characters "will be killed and transformed".
"We definitely kill people," she said. "It's safe to say that characters that people like will be killed. I find that usually when we kill somebody there's a certain amount of blowback because they're always characters that people like. I usually don't get thank you notes for the people that we kill."
Gamble added that characters from the show's past could reappear in the episode, titled 'The Man Who Knew Too Much'.
"We have that small group of characters that we go back to because they're precious," she explained. "The reason we go back to them again and again and again is because they were cool."
However, she dismissed rumours that the late John Winchester (Jeffrey Dean Morgan) will feature in the episode.
"You won't see John Winchester in season six," insisted Gamble. "[But] there's always a chance we'll bring him back at some point in the show."
The sixth season of Supernatural concludes tonight at 8/7c on The CW.
