The actress, who has previously worked with Whedon on Firefly and Angel, will reportedly play Bennett, a Dollhouse employee who shares a secret past with Echo.
Show creator Whedon has previously hinted that he was interested in casting Glau in the show's second season.
"I mentioned it to her before [Sarah Connor] was cancelled. I was like, 'You know, we should get you in the house'. But first we have to come up with something that works."
The upcoming second season was recently pushed back by Fox to Friday, September 25.
