The Hollywood Reporter confirms that Def has now finalised a deal to star in a multi-episode arc on the show's sixth season later this year.
The MC will portray an ex-convict who claims to have found God but comes under suspicion by Dexter (Michael C. Hall) after he continually winds up involved in violence.
Earlier this month, it was also confirmed that former Roswell star Colin Hanks had signed on to star in all 12 episodes of season six as a character called Travis.
Def has previously starred in a number of television series including House and The Boondocks, as well as having a role on HBO's upcoming comedy series Enlightened opposite Laura Dern.
Production on the new season of Dexter is due to begin next month for an expected launch this fall.
