Thomas will play Sandra Winthrop, a jewellery store owner, who has a thing for younger men. The actress will have a storyline, in which she is attracted to Eric Mabius's character Daniel Meade.
Last month, it was revealed that Dawson's Creek actor James Van Der Beek and Spice Girl Victoria Beckham would be appearing on the show.
The second season of Ugly Betty begins in the US on September 27.
