The show has been renewed by The CW for a 19th run, which will be a college-themed edition. Casting has already taken place.
However, Tyra Banks has confirmed that the trio will not be joining her on the new cycle.
"Fierce & love 2 my Nigel Barker, Mr Jay Manuel & Miss J," she tweeted. "ANTM turned us in2 a family. Excited for what the future holds for us. XOXO TyTy."
Barker also wrote about the news on his blog, saying: "After many fun-filled years with America's Next Top Model, I will not be returning to the programme for cycle 19."
The photographer, who has been on the judging panel since the beginning of the show, said that he will miss everyone involved and added that it will "always have a special place in [his] heart".
He continued: "This next chapter promises to be even more thrilling with the sky's the limit [sic]. I look forward to taking you with me."
Barker also hinted that he may have a new television project in the future, tweeting a fan: "You have 18 cycles of reruns! And just wait til my next show."
In a statement, the show's executive producer Ken Mok said that the trio had been an "integral" part of America's Next Top Model, adding: "They helped turn this show into the household name it is today.
"They have been amazing assets to the show and will always be a part of the Top Model family. We will continue to actively work with each of them on future projects."
Manuel has been the creative director for the America's Next Top Model photoshoots since the beginning of the show and hosted Canada's Next Top Model, while Alexander trains the models in their runway skills and also served as a judge for several cycles.
America's Next Top Model airs Wednesdays on The CW.
