Mitchell, who previously played Juliet Burke on the ABC drama, told E! Online that she would like to be reunited with her on-screen lover.
"I need that, right?" she asked. "Wouldn't that be wonderful?"
However, she admitted that a potential film career could hinder Holloway's chances of appearing on the programme.
"C'mon, he's a movie star," she said. "He needs to go be a movie star. He's so delightful."
The actress also insisted that the forthcoming second season of V will be "so much better" than the first.
"It comes back in November, and we just started filming," she explained. "I feel like [season one] set the groundwork and then it's off and running, which I needed it to be. It's more fun for me."
The new season of V is expected to begin airing in November.
